The great rune of the unborn will already be activated once you receive it, which means you do not need to do anything with it to unlock your power. There is a divine tower in Liurnia of the Lakes, but that area is a dungeon in itself, and it has nothing to do with the activation of the Great Rune you just had in your hands.

Instead, what it does is unlock the ability to respect your character completely when you speak with Rennala in the great Lucaria Raya Library.
